摘  要:为阐明静脉免疫球蛋白(IVIG)抑制新生儿 B 细胞免疫球蛋白释放的机制,采用间接免疫荧光标记法及 RT-PCR 法分别检测了 IVIG(5mg/ml)作用后的脐血淋巴细胞 CD40与 CD40配体表达及其分泌的 IL-2、IL-4 mRNA 表达。结果:1.IVIG 不改变脐血单个核细胞(CBMC)中CD40^+染色阳性细胞的百分比;IVIG促进了 CBMC 的 CD40L 表达;2.IVIG 抑制 CBMC IL-2、IL-4 mRNA 的表达。提示:IVIG 抑制 IL-2、IL-4mRNA 表达可能是 IVIG 抑制 B 细胞抗体释放的一重要机制。尽管 IVIG 可刺激体外 T 细胞 CD40L 表达的增加,但在 B 细胞上的 CD40及有关细胞因子(如 IL-2、IL-4)缺乏配合的情况下,CD40L 表达的增加可能无实际的意义。To determine the possible inhibitory mechanisms of intravenous immunoglobulin (IVIG) on the production of immunoglobulin in neonatal B cell,we examined the ability of IVIG (5mg/ml) to 1.regulate the number of CD40 and CD40 ligand positive cord blood lymphocytes,and 2. the expression of IL-2 and IL-4 mRNAs secreted by cord monocular cell (CBMC) using indirect immunofluorescent technique and the method of RT-PCR.The results showed that 1.IVIG had no effect upon the number of CD40~ cells in CBMC.IVIG induced an increase in the number of CD40L+ cells in primed CBMC.2.The addition of IVIG to the primed CBMC resulted in a significant inhibition of the expression of IL-2 and IL-4 mRNA.These results suggested that the IVIG-mediated decreased secretion of IL-2 and IL-4 at the mRNA level may be one of the inhibitory mechanisms of IVIG on the Ig production of cord blood B cells.THe expression of CD40L on cord blood primed T cells was increased by IVIG.However,in the absence of increased expression of CD40,co-stimulatory signals or other relevant cytokines,the increased level of CD40L expression confers no apparent advantage.
