摘  要:目的建立绝经期大鼠皮肤老化模型,探索皮肤自然老化的进程,为筛选抗皮肤老化的药物提供动物模型。方法2~3月龄Wister雌性大鼠随机分成手术组和非手术组,适应性喂养2周后,手术组大鼠切除双侧卵巢,再饲育50天[1]后处死大鼠,取皮肤组织做超氧化物歧化酶(SOD)、丙二醛(MDA)、羟脯氨酸(HYP)的测定及制作病理组织切片,观察大鼠皮肤的老化。结果与非手术组大鼠相比,手术组大鼠皮肤组织中SOD活性降低,MDA含量升高,HYP含量降低。大鼠皮肤组织病理切片显示与非手术组大鼠相比,手术组大鼠皮肤血管相对减少,表皮组织更加菲薄;真皮厚度变薄,胶原纤维减少,增粗老化,排列疏松,在真皮乳头层毛细血管减少;皮肤附属器,光镜下汗腺排列紊乱,汗腺数量减少,分泌细胞萎缩。结论双侧切除卵巢的生育期大鼠可以模拟皮肤自然老化。Objective To established the menopause aging skin model by ovariectomized of mice and select the drug to anti-skin aging to menopause mice. Methods Female Wister mice were randomly divided into ovariectomized or sham-operated groups, and biochemical analyses were carried out at 50 days after the operation. The dorsal skin samples were collected to determine super oxide dismutase(SOD) activity, malonaldehyde(MDA) and hydroryproline (HYP) content. The morphology of dorsal skins were observed and analyzed under the microscope. Results The content of MDA in the ovariectomy group significantly increased with statistical significance ( P 〈 0.05) , compared with the nomal control group. However, the activity of SOD and HYP content significantly raised which had statistical significance (P 〈 0.05). In contrast to the prepration group, thickness of dermis in control group showed significantly reduced and increased content of elastic fibers. Conclusion Ovariectomized mice can be established as the menopause aging skin model.
