摘 要:采用胶原酶消化法分离原代培养人网膜基质前脂肪细胞进行诱导分化。在诱导分化早期,前脂肪细胞因子1表达迅速降低,而脂蛋白脂酶、PPARγ/2表达迅速增加,CAAT/增强子结合蛋白α(c/EBPα)在成熟阶段表达丰度最高。瘦素mRNA表达和分泌随分化逐渐增高,抵抗素mRNA仅在前脂肪细胞表达和分泌而脂联素mRNA仅在成熟脂肪细胞表达和分泌,胰岛素样生长因子I(IGF-I)在前脂肪细胞诱导分化前后均有分泌。Stromal preadipocytes from human omental adipose tissues were isolated with enzyme-digesting method,and cultured for inducing differentiation. Preadipoeyte faetor-1 was abundant during preadipocyte stage, whereas lipoprotein lipase and PPAR-α2 mRNA were rapidly up-regulated at early stage of differentiation and CCAAT/enhancer binding protein expression was increased rather late. In addition, resistin expression and secretion was primarily detectable in preadipoeytes, leptin was gradually increased during adipogenesis, whereas adiponectin appeared specifically in mature adipocytes. Insulin-like growth factor-I secretion was remarkably detected in both preadipoeytes and mature adipocytes.
