摘  要:目的:研究AB-8树脂分离纯化玉米须总黄酮的工艺参数。方法:用紫外分光光度法测定玉米须总黄酮含量,以总黄酮吸附量、洗脱量、纯度及总黄酮收率为考察指标考查AB-8树脂对玉米须总黄酮的吸附行为。结果:确定适宜的上样条件为:玉米须上样液中总黄酮的浓度为1.88mg.mL-1,pH=6.0,上样流速为2.5BV/h;合适的洗脱条件为:90%的乙醇,流速为2.5BV/h。结论:用AB-8大孔吸附树脂对甘草黄酮有较好的吸附和解吸性能,纯化后玉米须总黄酮纯度为61.35%,总黄酮回收率为92.5%。Objective:To study the technique data for separation and purification the total flavonoids in the stigma maydis by AB-8 macroporous absorption resins.Methods:The concentration of flavones in liquorice was determined by ultraviolet spectrophotometry,and the absorption quantity of the total Flavonoids,the yield of the ethanol desorption materials,the content of obtained flavonoids and recovery rate of the Total Flavonoids were determined.Results:Optimal adsorption of flavones was achieved with total flavones concentration in the solution of 0.85mg/mL,the sample pH of 6.0,sample flow velocity of 2.5BV/h,and washing with 90%ethanol at the flow velocity of 2.5BV/h.Conclusion:AB-8 macroporous adsorption resin can be well applicable for enrichment of flavones in Stigma Maydis.The content of obtained flavonoids reached 61.35%,and the recovery rate reached 92.5%.
