摘 要:以槟榔叶片为植物材料,采用改良的SDS—CTAB方法提取其基因组DNA。结果表明,以水饱和酚代替Tris饱和酚对样品进行抽提的SDS—CTAB法可以获得纯度高(0D260/OD280在1.7~1.9、OD260/OD230大于2)、一级结构完整、分子量大于23kh的DNA,产率在50~120μg/g。该法所提DNA成功用于后续的分子生物学研究,并在花生、香蕉等植物上得到应用。The extraction of genomic DNA from the leaves of Areca catechu Linnaeus was researched by optimized SDS -CTAB method in order to obtain high - quality genomic DNA for subsequent molecular biological experiment. The results showed that high - quality DNA was isolated, the OD260/OD280 of the samples was between 1.7 and 1.9, the OD260/OD230 of the samples was more than 2, its primary structure was perfect, its molecular weight was more than 23 kb, and its yield was in range of 50 - 120μg/g. The DNA extracted by this method was successfully applied in the subsequent molecular biological experiment. The method was also applied to the extraction of DNA from peanut, banana and so on.
