摘 要:目的通过对蝙蝠呼肠孤病毒在BHK-21和Vero-E6细胞形态发生的比较研究,筛选出该病毒的最佳宿主细胞。方法将感染呼肠孤病毒XJV株的BHK-21和Vero-E6细胞用戊二醛、锇酸固定,制成超薄切片,用透射电镜观察。同时测定病毒滴度。结果XJV在2种细胞中的适应程度不同,XJV更适于在BHK-21细胞中增殖。XJV在2种细胞中的形态发生不同,在BHK-21细胞中形成的包涵体呈典型的晶格状排列,在Vero-E6细胞中形成的包涵体呈杂乱的球状。XJV用BHK-21和Vero-E6细胞育传5代,病毒滴度分别为105.5TCID50/ml和104.5TCID50/ml。结论BHK-21细胞是蝙蝠呼肠孤病毒XJV株的最佳宿主细胞。Objective The morphogenesis of an XJV strain in BHK 21and Vero-E6 cells was comparatively studied to select the best host cells. Methods BHK-21 and Vero-E6 cells were infected with the XJV strain and treated according to methods for preparation and observation of specimens using transmission electron microscopy. Results The adaptation of the XJV strain differed in two kinds of cells, and BHK-21 cells were more suitable for XJV strain proliferation. The morphogenesis of the XJV strain also differed in two kinds of cells. In the BHK-21 cells, the XJV inclusions were ar ranged in a crystal lattice, but forms in the Vero-E6 cells were disordered spheres. The viral titer in the BHK-21 cells was higher than in the Vero-E6 cells. Conclusion BHK 21 cells are the best host cells for XJV.
