摘 要:观察角质细胞生长因子对放疗和环磷酰胺引起胸腺萎缩的增殖作用。取正常BALB/C雄性小鼠,随机分为KGF处理组和对照组。KGF处理组和对照组均先给予4.5Gy钴射线照射,24h后分别连续皮下注射KGF(5mg/kg.d)和生理盐水7d。4周检测小鼠胸腺和脾脏组织的总细胞数和各T细胞亚群的细胞数;取正常BALB/C雄性小鼠,随机分为KGF处理组和对照组。KGF处理组和对照组均先连续腹腔注射环磷酰胺(300mg/kg)3d,注射完毕24h后再分别连续皮下注射KGF(5mg/kg·d)和生理盐水7d。2周后检测小鼠胸腺和脾脏组织的总细胞数和各T细胞亚群的细胞数。结果:细胞计数和流式细胞仪检测显示,分别经放疗和环磷酰胺处理的小鼠,与对照组相比,KGF处理组平均胸腺细胞总数和CD4SP、CD8SP、DP、DN各T细胞亚群的平均细胞数以及KGF处理组平均脾脏细胞总数和CD4SP、CD8SP各T细胞亚群的平均细胞数均有明显增加(P<0.05)。角质细胞生长因子对放疗和环磷酰胺引起的胸腺萎缩均具有增殖作用,同时可以增加成熟T细胞向脾脏的输出而增强机体的免疫功能。To observe the enhancement effect of keratinocyte growth factor (KGF) on the proliferation of atrophy thymus caused by radiotherapy and cyclophosphamide respectively, male BALB/C mice were randomly divided into 4 groups (2 KGF test groups and 2 control groups). 2 goup mice (1 test and 1 control) and other 2 groups (1 test and 1 contol) were exposed in Co ray of 4. 5 Gy and intraperitoneally injected with eyclophosphamide (300 mg/kg) for 3 consecutiee days respectively to make atrophy thymus model animal. 2 KGF group mice and 2 control groups were subcutaneously injected with KGF (5 mg/kg.d) and saline respectively for 7 consecutive days. The cell number of thymus and spleen and T cell subsets number were detected. The total cell number and T cell subsets number of thymus and spleen were detected 2 weeks later. Cell counting and flow cytometry showed that both in radiotherapy-treated mice and in cyclophosphamide-treated mice, the mean number of absolute cell and CD4SP, CD8SP, DP, DN T cell subsets of thymus and the mean number of absolute cell and CD4SP, CD8SP T cell subsets of spleen increased significantly in KGF- treated group as compared with the control group (P〈0. 05). KGF could enhance the proliferation of atrophy thymus caused by radiotherapy and by cyclophosphamide respectively and increase the peripheral output of mature T cells to spleen thus strengthening the body immune function.
