抗衰益智胶囊对阿尔茨海默病模型大鼠学习记忆能力、GSH-Px及脑组织超微结构的影响

摘　　要：目的观察抗衰益智胶囊对阿尔茨海默病模型大鼠学习记忆能力、脑组织谷胱甘肽过氧化物酶(GSH-Px)活性及脑组织结构的影响。方法采用D-半乳糖致亚急性衰老合并脑内注射喹啉酸复制阿尔茨海默病大鼠模型。检测抗衰益智胶囊对模型大鼠脑组织GSH-Px活性及脑组织超微结构的影响,并采用跳台法和Morris水迷宫实验观察抗衰益智胶囊对大鼠学习记忆能力的保护作用。结果抗衰益智胶囊能够改善模型大鼠的学习记忆成绩,包括缩短模型大鼠在跳台中寻找安全平台的逃避潜伏期(P<0.01)和在Morris水迷宫实验中的5 d平均潜伏期(P<0.01),增加安全平台停留时间(P<0.05);能明显升高模型大鼠脑组织GSH-Px水平(P<0.01)。结论抗衰益智胶囊能提高机体脑组织中GSH-Px活力,清除氧自由基,减少脂质过氧化反应,减少神经元的损伤,从而起到保护脑细胞的作用;对阿尔茨海默病模型大鼠的学习记忆能力有保护作用。Objective To study the effects of the Kangshuaiyizhi capsule （KSYZ） on the ability of learning and memory, the activity of glutathion peroxidase （GSH-Px） and brain tissue ultrastructure in rat model of Alzheimer＇s disease induced by D-galactose and quinoline acid. Methods The AD model of rats was established by injecting D-galactose and quinoline acid. Then the effect of KSYZ on the activity of GSH-Px and brain tissue ultrastructure, learning and memory retention disorder in the model rats were studied by the test of Morris watermaze and by the measure of step-down test. Results Compared with model group, KSYZ could obviously improve the ability of learning and memory, for example, significantly shorten the escape delitescence （P 〈0.01） of searching safety platform in the step-down test and the five days＇ average delitescence （P 〈0.01） in Morris water maze test, and increase the detention time of safety platform （P 〈0.05）. It could also improve the activity of GSH-Px in brain tissue of rats （P 〈0.01）. Conclusion It suggests that KSYZ has functions of improving body＇s resisting oxidation and eliminating free radicle, reducing injure to nerve-cell and improving the ability of learning and memory. Thus KSYZ offers protection to the brain tissue of Alzheimer＇s disease rats.
