摘 要:选取分离自大洋、深海的28株酵母和143株霉菌,采用在培养基中添加尼罗红比较表观荧光强度的方法来筛选出酵母2株和霉菌7株,但菌体呈红色的酵母(17株)及菌丝为黑色的霉菌(28株)不适用于此法;通过单位OD600荧光强度的高低筛选出1株菌落为微红色的酵母,产油性能好于用第一种方法挑选出的2株酵母,且菌体颜色对结果并无影响,但只适合筛选酵母.通过上述2种方法筛选出高油脂菌株,大多来源于深海.Two methods for rapid screening intracellular lipid content in oceanic oleaginous microorganisms (28 yeasts and 143 moulds) were established using a fluorescent probe Nile red in this paper. The first method screened oceanic oleaginous microorganisms using Nile red, established by the Nile red direcdy included in the medium and then compared the fluorescence intensity under ultraviolet. 2 white yeasts out of 28 yeasts and 7 non - black moulds out of 143 moulds were prominent in producing intracellular fatty acid. However, this method was not suitable for red yeasts(17) and black moulds(28). The second method, by comparing the fluorescence intensity per OD600, one reddish yeast was selected with much higher fluorescence intensity. However, this method could not apply to moulds due to their inter - twisted mycelia. With above methods most of screened microorganisms capable of producing high content intraceltular fatty acid are from deep sea.
