摘  要:目的探讨慢性期慢性粒细胞白血病(CML)患者骨髓间质干细胞(BMMSCs)体外生长特性和核转化因子κB(NF-κB)mRNA表达的变化及意义。方法选择5例慢性期CML患者为A组和6例健康人为B组,取骨髓单个核细胞行BMMSCs体外培养,观察BMMSCs体外生长形态并计数,绘制生长曲线。取第3、4代BMMSCs提取总RNA、逆转录RT-PCR检测NF-κB mRNA表达量。结果CML患者BMMSCs生长活性较对照组无明显差别,但其NF-κB mRNA表达量较对照组升高(P<0.05)。结论CML患者BMMSCs中NF-κB基因高表达影响CML发病。Objective To investigate the expression and significance of nuclear factor (NF)-κB gene in bone marrow mesenchymal stem cells(BMMSCs) derived from patients with chronic myeloid leukemia(CML). Methods BMMSCs of 5 CML patients and 6 healthy persons were cultured in vitro. The morphology of BMMSCs was observed and the growth curve was portrayed according to the daily results of BM MSCs proliferation. Total RNA was extracted from third and fourth generation of BM MSCs and the expression of NF-κB gene of BM MSCs was measured by reverse transcription-PCR. Results The growth activity of BMMSCs was not significantly different between two groups,but the expression of NF-κB gene of BMMSCs was higher in group A than that in group B (P〈0. 05). Conclusion The abnormally high expression of NF-κB of BMMSCs takes part in the pathogenesis in patients with CML.
