热休克蛋白60在子宫颈癌组织中的表达及其临床意义

摘　　要：目的研究热休克蛋白60(heat shock protein,HSP60)在子宫颈癌中的表达及意义。方法应用半定量逆转录聚合酶链反应(RT-PCR)和免疫印迹分析法检测子宫颈癌手术切除组织57例,正常子宫颈组织57例,观察热休克蛋白60的表达与子宫颈癌发生发展的关系。结果①正常子宫颈组织HSP60 mRNA表达明显少于子宫颈癌组织(t=2.65,P<0.05)。②57例子宫颈癌组织和57例正常子宫颈组织中,HSP60的表达阳性检出率分别为:82.5%、19.3%;HSP60的过表达率为57.9%、7.0%。③HSP60在子宫颈癌组织的不同分期中表达差异无显著性(P>0.05)。④子宫颈正常组织HSP60蛋白的表达水平明显低于子宫颈癌组织(t=3.132,P<0.05)。结论HSP60的表达与子宫颈癌组织的发生密切相关。HSP60的表达与子宫颈癌组织的不同分期无关。Objective To investigate the expression of heat shock protein （HSP） 60 and analyze the significance in cervical carcinoma. Methods The cervical tissue samples of 57 cases of cervical cancer and 57 normal cervical tissues were collected. The expression of HSP60 were detected by semi-quantitative reverse transcription polymerase chain reaction （RT-PCR）. The protein expressions of HSP60 were determined by Western blot analysis. To observe the relationship between HSP60 expression and the occurrence and development of cervical cancer. Results HSP60 mRNA expression in normal cervical tissue was significantly lesser than that in cervical cancer tissue （ t = 2.65, P 〈 0.05）. The positive rate of HSP60 expression in cervical cancer and normal cervix was 82.5% and 19.3% respectively. The over-expression rate of HSP60 was 57.9% and 7.0% respectively. The difference of HSP60 expression had no significance at different stages of cervical cancer tissue （P 〉 0. 05 ）. The levels of HSP60 protein expression in normal cervical tissue was significantly lower than the expression in cervical cancer tissue （t = 3. 132, P 〈0.05）. Conclusion The expression of HSP60 is closely related to the incidence of cervical cancer, but there is no relationship with different stages of cervical cancer.
