摘  要:目的根据羟自由基与水杨酸(SA)氧化反应的动力学过程,建立动力学分光光度法测定中药多糖对羟自由基(.OH)的清除率。方法应用紫外分光光度法,建立.OH与SA反应的动力学曲线,根据动力学曲线斜率的变化测定中药陈皮和灵芝多糖对.OH的清除率,并用维生素C进行方法的重复性与可靠性验证。结果测定陈皮多糖对.OH的50%清除率(IC50)为232.6 mg.L-1;灵芝IC50为0.856 1 g.L-1;维生素C的IC50为0.254 4 mmol.L-1,RSD为0.2%(n=5)。结论此方法灵敏可靠,可用于中药多糖成分对.OH清除率的测定。OBJECTIVE The reaction dynamic of hydroxyl free radical(.OH) and salicylic acid was explored,the elimination ratios of polysaccharide extractive of Chinese traditional medicine to.OH was determined by kinetic spectrophotometry.METHODS The dynamics curve of the reaction.OH and salicylic acid was built by UV method.The elimination ratio of tangerine pee polysaccharide and ganoderma lucidum polysaccharide to.OH was detemined based on the slope change of the graph.Repetitiveness and reliability of this method was tested by vitamin C.RESULTS The 50% elimination ratio(IC50) of polysaccharide extractive of tangerine pee and ganoderma lucidum to.OH was 232.6 mg.L-1 and 0.856 1 g.L-1 respectively,the IC50 of vitamin C was 0.254 4 mmol.L-1,RSD was 0.2%(n=5).CONCLUSION The methodis sensitive,reliable,and it is suitable to be used to determine the elimination ratio of polysaccharide extractive of Chinese traditional medicine to.OH.
