Influence of Chrysanthemum indium on collagen accumulation and signaling transduction pathways in left ventricular tissue of cardiac hypertrophy in rats

摘  要:目的:研究野菊花水提液对压力负荷大鼠心肌组织胶原及信号传导的影响。方法:腹主动脉不完全结扎法(ab-dominal aortic banding,AAB)制作大鼠心肌肥厚、心室重构模型。35 d药物干预后,测量大鼠血压和心脏质量指数,取左室心肌病理切片,经HE染色测量心肌细胞横断面面积(myocardium cell cross section,MCCS);经苦味酸天狼猩红染色测量心肌组织胶原容积分数(collagen volume fraction,CVF),测量心肌血管周围胶原面积(perivascular collagen area,PVCA)和Ⅰ,Ⅲ型胶原的含量;免疫组化法测量心肌组织PKC,bFGF,P38的含量。结果:与假手术组比较,模型组动物心脏指数和心肌细胞横断面面积明显变大,血压升高,心肌组织CVF,PVCA,Ⅰ,Ⅲ型胶原生成增加,PKC,bFGF和P38表达量显著增多(P<0.05)。野菊花水提液能明显降低心脏指数和血压,缩小心肌细胞横截面面积,减少PVCA,CVF,Ⅰ,Ⅲ型胶原,降低PKC,bFGF和P38的表达(P<0.05)。结论:野菊花水提液具有减少心肌胶原沉积,抗实验性心室重构的作用,其作用机制与降低心脏的负荷,调节信号传导有关。Objective: To evaluate the influence of Chrysanthemum indium on collagen accumulation and signaling transduetion pathways in left ventricle tissue of cardiac hypertrophy induced by abdominal aortic banding in rats. Method : Ventricular remodeling was induced by abdominal aortic banding( AAB) in rats. After 35 day treatment, the blood pressure was measured, then the ratios of LVW/BW and HW/BW were caleulated. The histological assay was performed by HE staining for determining the myocardium cell cross section and picric acid/sirius red staining for determining collagen content. Immunohistochemistry was used to detect the protein expressions of PKC, bFGF and P38. Result: The experimental data demonstrated that C. indium could decrease blood pressure and the cardiac indexes of LVW/BW and HW/BW, significantly diminish cross sectional area of cardiomyocyte, ameliorate collagen accti- mulation such as collagen volume fraction, perivascular collagen area and collagen distributions of type Ⅰ and Ⅲ and significantly down regulate the protein expressions of PKC, bFGF and P38 (P 〈 0. 05). Conclusion:C. indium ean significantly attenuate the experimental ventricular remodeling. The mechanism may be related to reducing the blood pressure, decreasing the total collagen content of left ventricle tissue and modulating signaling transduction pathway.
