摘  要:目的:观察补肾通脉方对多囊卵巢综合征(polycystic ovary syndrome,PCOS)的胰岛素抵抗(insulin resistance,IR)大鼠的靶器官中胰岛素受体底物-1丝氨酸307(insulin receptor substrate-1 Ser307,IRS-1 Ser307)磷酸化表达的影响,探讨补肾通脉方改善PCOS的IR的机制。方法:建立IR的PCOS大鼠模型,成模大鼠随机分为模型组和中药组,另设13只正常对照组。中药组大鼠以补肾通脉方干预。各组动情后期大鼠于门静脉推注胰岛素前后各处死一半。免疫印迹法(Western blot)检测各组大鼠肝脏、脂肪IRS-1 Ser307磷酸化蛋白表达,免疫组化法检测卵巢IRS-1 Ser307磷酸化蛋白表达。结果:模型组IRS-1 Ser307磷酸化在肝脏、脂肪和卵巢中表达均明显高于正常组(P<0.05,P<0.01),中药组均显著低于模型组(P<0.05,P<0.01)。各组肝脏、脂肪和卵巢中IRS-1 Ser307磷酸化表达在胰岛素刺激后均较刺激前明显增多(P<0.05)。结论:补肾通脉方可下调胰岛素靶组织IRS-1 Ser307磷酸化水平、改善胰岛素信号转导,这可能是其改善PCOS的IR的机制之一。Objective: To investigate the effects of Bushen Tongmai recipe on the phospharylation expression of insulin receptor substrate-1 Set307 (IRS-1 Ser307 ) in insulin target tissues of polycystic ovarian syndrome (PCOS) rats accompanying with insulin resistance (IR). Method: Rats of model of PCOS accompanying with IR were randomly divided into the model group and the traditional Chinese medicine(TCM) group. Meanwhile, a group of 13 rats of the same age was considered as the normal control group. The TCM group was administered with BSTMR. Half of each group was given insulin injection through portal vein in late estrus. Another half didnt receive insulin injection. The phospharylation levels of IRS-1 Ser307 in liver and fatty tissues were measured by Western blot. The phospharylation level of IRS-1 Ser307 in ovary tissue was measured by immunohistochemistry. Result: The phospharylation expressions of IRS-I Ser307 in liver, fat and ovary in the model group were higher than those in the normal group (P 〈 0. 05, P 〈 0. 01 ) ; those criteria in the TCM group were lower than those in the model group(P 〈 0. 05 ). After insulin stimulation, the phospharylation expressions of IRS-1 Set307 were higher than those before insulin stimulation (P 〈 0. 05). Conclusion: Bushen Tongmai recipe could attenuate the phospharylation expression of IRS-1 Ser307 and then enhance insulin signal transduction, which may be one of the mechanisms of Bushen Tongmai recipe in improving PCOS accompanying with IR.
