摘 要:目的:探讨纳米化对乙型肝炎免疫球蛋白(HBIG)的细胞内浓度和渗透率的影响。方法:取HBIG聚氰基丙烯酸正丁酯纳米粒(HBIG-PBCA-NP),以QSG7701细胞为体外细胞模型,首先采用MTT法检测HBIG-PBCA-NP和HBIG在试验浓度下对细胞的毒性作用;再分别对细胞加入HBIG-PBCA-NP和HBIG处理后HBIG的胞内浓度和HBIG渗透率进行不同时间点的对比考察。结果:HBIG-PBCA-NP和HBIG在试验浓度下对细胞无毒性作用;HBIG-PBCA-NP组的HBIG胞内浓度和HBIG渗透率在各时间点均明显高于HBIG组(P<0.05或P<0.01)。结论:HBIG纳米化能增强HBIG的细胞渗透能力,提高HBIG的胞内浓度。OBJECTIVE: To investigate the effect of nanominiaturization on concentration and penetration of hepatitis B im- mune globulin (HBIG) in cells. METHODS: HBIG polybutylcyano-acrylate nanoparticles (HBIG-PBCA-NP) were prepared and QSG7701 cell lines were used as in vitro cell model. First, MTT method was carried out to determine HBIG-PBCA-NP and HBIG' s toxic action to cells under the test concentration. Then HBIG-PBCA-NP and HBIG were added into cells to compare intracellular concentrations of HBIG and penetrability of HBIG at different time. RESULTS: It was found that HBIG-PBCA-NP and HBIG had no toxic action to cells and all of determined cellular HBIG concentration in the HBIG-PBCA-NP group was higher than in HBIG group (P〈0.05 or P〈0.01). CONCLUSION: The results indicate that penetrability and concentration of HBIG across the cellular membrane has been improved by nanominiaturization.
