摘 要:目的探讨葡萄糖和晚期糖基化终产物(AGE)联合刺激对人单核细胞THP-1血红素加氧酶1(HO_1)表达的影响。方法分别以15mmol/L葡萄糖、100μg/ml AGE和二者联合刺激THP-1,比较各组活性氧(ROS)产量,培养液丙二醛(MDA)、肿瘤坏死因子α(TNFα)水平和细胞HO-lmRNA及蛋白的表达量。结果葡萄糖和AGE联合刺激的ROS产量、MDA、TNFα水平及HO-1mRNA和蛋白表达均明显高于单独刺激组。结论葡萄糖和AGE联合刺激可导致THP-1细胞氧化应激和HO-1表达增加。Objective To investigate in effects of high glucose and advanced glycation end products (AGEs) on expressions of heine oxygenase-I in THP-1 cells. Methods THP-1 cells were incubated seperatatedly with 15 mmol/L glucose, 100 μg/ml AGEs and combination of the above two agents. The levels of ROS, malondialdehyde (MDA) and tumor necrosis factor α (TNFα) were determined after stimulating for 24h. Also the expressions of HO-1 mRNA and protein were determined seperatedly by RT- PCR and Western blotting. Results As compared with the simple glucose or AGEs groups, the 15 mmol/L glucose + 100 μg/ml AGEs group had the highest output of ROS(107. 21 ±8. 94), MDA (3. 26+0.32 nmol/ml), TNFα(38. 09±14. 97 pg/ml) and the expression of HO-1. Conclusions When incubated by high glucose plus AGEs, the oxidative stress and expression of HO-1 in THP-1 cell are augmented significantly. HO-1 may be a signal of oxidative stress in diabetes mellitus and the induction of HO-1 may be a new method of the treatment of diabetic complication.
