摘 要:目的:探讨冠心宁注射液对大鼠缺血再灌注心肌的影响。方法:40只SD大鼠被随机分为四组,每组10只,分别为:假手术组、梗塞对照组、冠心宁注射液低剂量组、冠心宁注射液高剂量组。采用结扎冠状动脉左前降支的方法复制急性心肌缺血再灌注模型,观察大鼠急性心肌梗塞范围、梗塞心肌组织细胞因子白细胞介素(IL)-10、肿瘤坏死因子(TNF)-α的水平。结果:(1)冠心宁注射液低、高剂量组心肌梗塞范围均显著小于梗塞对照组(P<0.01),两剂量组之间无显著差异(P>0.05);(2)冠心宁注射液低、高剂量组IL-10水平高于梗塞对照组(P<0.05),低于假手术组(P<0.01);TNF-α水平低于梗塞对照组(P<0.05),高于假手术组(P<0.01),而两剂量组之间无显著差异(P均>0.05)。结论:冠心宁注射液能抑制大鼠坏死心肌TNF-α的表达,促进IL-10的生成,抑制炎症反应,减少心肌梗塞范围,具有较好的心肌保护作用。Objective: To study the protective effect of Guanxinning injection on myocardial ischemia/reperfusion injury in rats. Methods: A total of 40 SD rats were randomly divided into four groups, each with 10 rats (including two rats were used in paintings and the other eight used in experiment observation from each group). Four groups were as follows: sham operation group, the infarction control group, Guanxinning injection of low-dose group, Guanxinning in- jection of high-dose group. The model of acute myocardial ischemia rat was obtained by legating the left anterior descending coronary artery. Results: The extent of myocardial infraction of Guanxinning injection high and low dosage treated group reduced significantly compared with that of model group (P〈0. 01), and two dosage treated group was not different (P〉0. 1) . The IL-10 levels in necrotic myocardial cell after giving Guanxinning injection were higher than that in the I/R group (P〈0. 05), and were lower than that in sham operation group (P〈0. 01). While the TNF-α levels in necrotic myocardial cell after giving Guanxinning injection were lower than that in I/R group (P〈 0.05), and were higher in the sham operation group (P〈0. 01). The levels of IL-10 and TNF-α in necrotic myocardial cell in Guanxinning injection high, low dosage group were not singificant difference (P:〉0. 05). Conclusion.. Guanxinning injection can reduce TNF-α and increase IL-10 levels in the necrotic myocardial cell, decrease the extent of myocardial infraction, and has protective effects on myocardial ischemia/reperfusion injury in rats.
