摘  要:用带嵌合基因35S-Ac-GUS的农杆菌双元载体转化普通烟草品种Nc89,对转基因植株(T0)及其回交后代(BCF1)进行GUS组织化学分析,检测到转座因子Ac在部分T0代和BCF1代植株中转座。用Ac作探针,与上述T0代和BCF1代植株的基因组DNA进行Southern杂交,结果表明,Ac从原来所在的T-DNA解离,并整合到烟草基因组的其它位点。同时对Ac-2植株T1代的NPTII基因和Ac因子进行多重PCR分析,发现Ac因子与NPTI基因间出现遗传重组,也间接证明Ac发生了转座。最后还对转座因子标签研究中转座基因型及突变体筛选的策略进行了讨论。The maize transposable element Activator was transformed into Nicotiana tabacum L. with Agrobacterium tumefaciens harboring the chimeric gene 35S Ac GUS. Transgenic plants and their offsprings were examined by GUS (β glucuronidase) histochemical assay. Different transposition patterns of Ac element were observed in the tobacco somatic tissues. Multiple PCR with selfed progenies of the transgenic plant Ac 2 revealed that genetic recombination occurred between Ac and NPTII gene (including T DNA). The genomic DNAs of transgenic plants and their offsprings were probed with Ac and GUS fragments in Southern blot analysis. The results indicated that Ac transposed in T 0 and T 1 generations. A strategy for the selection of transposition genotypes was discussed.
