摘  要:[目的]确定穿龙薯蓣花粉活力及柱头可授性的快速、有效测定方法。[方法]分别采用I2-KI染色法、TTC染色法、亚甲基蓝染色法、MTT染色法和无机盐培养法测定穿龙薯蓣的花粉活力,采用联苯胺-过氧化氢法和MTT染色法测定其柱头的可授性。[结果]I2-KI和TTC染色法对穿龙薯蓣花粉的染色效果不理想,不适于观测其花粉活力;亚甲基蓝染色法使花粉生活力的测定值偏高;MTT染色法使花粉细胞出现紫色、淡红色、无色3种颜色反应,利于分辨花粉生活力的有无。联苯胺-过氧化氢法不适于观测穿龙薯蓣柱头的可授性;MTT染色法能较准确地观测出不同时期柱头可授性的动态变化。[结论]MTT染色法适于检测穿龙薯蓣的花粉活力及柱头可授性。[Objective] The study was to determine a fast and effective determination method of pollen viability and stigma receptivity of Dioscorea nipponica Mak.[Method] Staining methods with I2-KI,TTC,methylene blue and MTT and inorganic salt culture method were taken to determine the pollen viability of D.nipponica,and the methods of benzidine-H2O2 and MTT staining were taken to determine its stigma receptivity.[Result] The staining effects of I2-KI and TTC on pollen of D.nipponica were not ideal,so the two methods were not suitable for observing the pollen viability of D.nipponica.Methylene blue staining made the measured value of pollen viability on the high side.MTT staining made pollen cell occur three kinds of color reaction of purple,faint red and colorless,which was beneficial to identify pollen viability.Benzidine-H2O2 method was not suitable for observing the stigma receptivity of D.nipponica.The dynamic changes of stigma receptivity in different periods could be accurately observed with MTT staining method.[Conclusion] MTT staining method was suitable for detecting the pollen viability and stigma receptivity of D.nipponica.
