摘 要:研究WR2721(氨基硫醇类药)对放烧复合伤的预防效果及其对造血功能的保护作用。结果证明:WR2721能明显提高放烧小鼠的活存率,并延长死亡动物的活存时间。用WR2721预防后,放烧小鼠的LD50由对照组的5.19Gy增大到9.53Gy,DRF为1.84。WR2721对放烧小鼠的预防作用以200mg/kg照射前立即给药效果最好。放烧小鼠用WR2721预防后,与对照组相比,外周血白细胞数和骨髓有核细胞数下降幅度少,恢复快,骨髓粒单系祖细胞伤后第1d和7d及骨髓造血干细胞伤后第7d和14d明显高于对照组。说明,WR2721对放烧小鼠的预防作用,至少部分是通过保护造血功能而实现的。Protective effect of agent WR 2721 on hematopoiesis and prophylaxis for the combined radiation burn injury were investigated in the present study. It was found that pretreatment with WR 2721 could significantly raised the survival rate and suwival time of mice with the combined radiation burn injury. Prophylactic use of WR 2721 resulted in an increase of LD50 value from 5.19 Gy of the control to 9.53 Gy of the pretreatment. DRF (dose reduction factor) was 1.84. Prophylactic effect of WR 2721 was gained by the immediate use before irradiation with the dose of 200 mg/kg. Pretreatment of WR 2721 in mice with the combined injury brought about a lesser decrease in peripheral WBC count and an earlier recovery compared with the control. Bone marrow myelocyte and monocyte counts were markedly higher than the control at day 1 and 7 post injury and bone marrow hematopoietic stem cell count was significantly higher than the control at day 7 and 14. Prophylactic effect of WR 2721 on the combined radiation burn injury was at least partially due to its protective effect on hematopoiesis.
