摘  要:[目的]研究藻蓝蛋白对糖尿病大鼠胰岛细胞功能的影响及其可能的机制。[方法]健康雄性Wistar大鼠32只,应用高脂高糖饲料喂养联合小剂量链脲佐菌素(STZ)腹腔注射方法建立2型糖尿病大鼠模型,藻蓝蛋白、二甲双胍灌胃治疗,免疫组化技术检测胰岛细胞中核因子κB(NFκB)和NFкB抑制蛋白(IκB)的表达。[结果]大鼠造模成功后空腹血糖明显升高,经干预治疗后,藻蓝蛋白组、二甲双胍组大鼠空腹血糖比模型组显著降低(P<0.05)。糖尿病大鼠胰岛细胞NFκB表达明显增高,IκB表达明显下降。经藻蓝蛋白和二甲双胍治疗后,大鼠胰岛细胞NFκB表达较糖尿病模型组均明显降低,而IκB表达较糖尿病组显著增强(P<0.05)。[结论]藻蓝蛋白可抑制2型糖尿病大鼠胰岛细胞IκB/NFκB通路的活性,对胰岛细胞具有一定的保护作用。[Objective] To investigate the effect and action mechanism of phycocyanin on the function of islet cells of rats with diabetic mellitus. [Methods] A model of Type 2 diabetes mellitus was induced by high fat diet and intraperitoneal injection of streptozotocin (STZ) in low dose in 32 healthy male Wistar rats. They were treated with phyeocyanin and dimethylbiguanide. The expression of nuclear factors B (NFKB) and inhibitory KB (IKB) in islet cells was detected by immunohistochemistry. [Results] The free blood glucose (FBG) in model group was significantly higher than that in rats of normal control group, and was significantly lower in treated groups compared with model group (P〈0.05) .The expressions of NFKB protein increased while IKB protein decreased significantly in islet cells of model group rats. After treatment with phycocyanin and dimethylbiguanide, the NFKB protein expressed lowly and IKB protein expressed highly than those in model group rats. (P〈0.05). [Conclusion] It is suggested that phyeoeyanin may inhibit the activity of IKB/NFKB pathway to protect the islet cells of diabetic mellitus in rats.
