摘  要:利用电镜细胞化学的方法,对烟草进入旺长期正常生长条件和干旱胁迫下叶片细胞壁糖蛋白(expansin)的积累动态变化进行了观察。结果表明:(1)处理第16 d,正常生长条件下可以观察到糖蛋白紧紧附着在细胞壁两侧;干旱条件下只有少量糖蛋白附着于细胞壁两侧;(2)处理第28 d,正常生长条件下,附着在叶肉细胞壁的糖蛋白层继续增厚,且连成一片;干旱处理条件下叶肉细胞壁几乎没有糖蛋白附着,大部分已经脱落于细胞间隙;(3)处理第40 d,正常条件下叶片细胞壁表面附着的糖蛋白大量脱落,干旱条件下叶肉细胞壁基本没有糖蛋白附着;(4)与正常生长的烟株形态相比,干旱处理后烟株生长变慢,叶面积变小,株型紧凑,叶片不开展,表明细胞壁糖蛋白的积累与烟株生长、叶面积扩展有密切关系,同时在烟株初期的抗逆性中也起到一定的作用。Accumulation status of cytoderm expansin in flue-cured tobacco leaf was studied by electron microscope under drought and normal condition during vigorous growing period. Results showed that: (1) After treating 16 days, under normal condition, expansin was close to cytoderm, while little cytoderm expansin attached to diachyma cytoderm under drought stress; (2) After treating 28 days, under normal condition, the expansin layer increased continuously and joined together, while un- der drought stress, almost nothing attached to the diachyma cytoderm, mostly dropped off intercellular space; (3) After treat- ing 40 days, cytoderm expansin fell off under normal condition, and almost nothing attached to the diachyma cytoderm under drought stress; (4) Compared with tobacco morphology under normal condition, growing rate under drought slowed down, leaf area diminished, plant types inseparably closed, leaves were unfold. There was close relationship between cytoderm expansin accumulation and change of leaf area, and hence affected tobacco drought resistance.
