蒿芩清胆汤和玉屏风散对流感病毒感染湿热证小鼠T淋巴细胞亚群和血清Th1/Th2细胞因子变化的影响

摘　　要：目的:应用蒿芩清胆汤和玉屏风散对流感病毒感染湿热证模型进行治疗,比较用药前后小鼠T淋巴细胞亚群和Th1/Th2细胞平衡的变化,初步探讨两种中药方法各自对改善小鼠免疫功能的特点,为两者更深一层差异的研究打下理论基础。方法:将75只小鼠随机分为正常组、模型组、阳性药物组、蒿芩清胆汤组和玉屏风组。造模成功后分别用流式细胞仪测定T淋巴细胞亚群(CD4+、CD8+)的浓度,用双抗体夹心ELISA法和酶标仪测定IL-4/IFN-rOD值,并通过换算得出其浓度。结果:与模型组相比,两组中药方剂均能升高CD3+CD4+/CD3+CD8+(P=0.001,0.04),且玉屏风散比蒿芩清胆汤效果略明显;而在Th1/Th2细胞平衡中,两种中药方剂均能降低IL-4/IFN-r浓度(P=0.006;P=0.03),蒿芩清胆汤较玉屏风散作用显著。结论:两种中药方法均能改善模型小鼠T淋巴细胞亚群比值和Th1/Th2细胞平衡,但效果有所不同,初步认为两者可能在对T淋巴细胞亚群比值和Th1/Th2细胞平衡的信号转导上存在着一定的差异,且有待于进一步的研究。Objective：Hao Qin Qing Dan decotion and Jade Screen Powder used in treatments on the mouse models of Damp-heat syndrome caught by infecting H1N1,to compare the changes of T-lymphocyte subsets and serum Th1/Th2 cell factors in the mouse between before-medication and after-medication,then Preliminarily explore the ways of each Chinese Medicine in improving the Immune Functions of the mouse models and make a theoretical basis for researching their further differences.Methods：Randomly divided 75 mouses into five groups：control group,model group,Virazole group,Hao Qin Qing Dan decotion group and Jade Screen Powder group.After establishing the model,we analyzed the concentration of T-lymphocyte subsets by flow cytometer,ODs of the IL-4 and IFN-r by Double-antibody sandwich ELISA method and the Microplate reader,then obtained the concentrations of IL-4 and IFN-r by Conversion formula.Results：Compared with the model one,Both Hao Qin Qing Dan decotion group and Jade Screen Powder group were able to raise the ratios of CD＋3 CD＋4/ CD＋3 CD＋8,especially the Jade Screen Powder one. Both of these two groups can reduce the ratio of the IL-4/IFN-r and the Hao Qin Qing Dan decotion group showed more effective.Conclusion：Both Hao Qin Qing Dan decotion and Jade Screen Powder can improve the ratio of T-lymphocyte subsets and serum Th1/Th2 cell factors.May be there are something differences in the signal transduction between them and further research is necessary to be carried out.
