摘  要:目的观察大鼠急性心肌梗死早期肝细胞生长因子(HGF)基因表达的变化。方法SD大鼠随机分为对照组与急性心肌梗死(AMI)组:结扎左冠状动脉前降支,操作结束后1、2、4、6、12h处死大鼠。应用伊文思蓝及氯化三苯硝基四氮唑红(TTC)染色,测定心肌梗死范围;应用RT-PCR测定各组心肌缺血区HGF基因动态表达情况。结果冠状动脉左前降支结扎术后20min内心电图ST段、T波进行性抬高;左心室心肌经伊文思蓝和TTC染色后见蓝色正常区、砖红色缺血区和灰白色坏死区;缺血12h称量梗死心肌范围占危险区心肌重量百分比为(66.44±19.24)%。心肌梗死后随时间延长HGFmRNA表达逐渐上升(P<0.05)。结论HGF基因水平增高参与急性心肌梗死早期心肌组织损伤。Objective To investigate the expression of hepatocyte growth factor(HGF) mRNA in early period in rats with acute myocardial infarction(AMl). Methods The SD rats were randomly divided into control group and AMI group. RT-PCR was used to investigate the expression levels of HGF in rats with AMI at 1 h,2h ,4h, 6h and 12h after AMI. Results The gene expression of HGF was significantly increased in AMI group at 4h, at 6h,at 12h,as compared with in control group( P 〈0.05 ). Conclusion The increase of the levels of HGF gene expression may be involed in the injury of myocardium at early stage of AMI.
