hURAT1基因5'UTR区荧光素酶表达重组体的构建

摘　　要：目的构建人尿酸盐转运蛋白1(hURAT1)基因5’UTR区荧光素酶表达重组体。方法PCR扩增包含hURAT1基因启动子最小功能单位和5’UTR区域在内的DNA片段,克隆至荧光素酶表达载体pGL3-basic,构建hURAT1 5’UTR区表达重组体。重组体经双酶切后测序进行鉴定。结果克隆获得的590 bp的DNA序列与GenBank报道的一致,且插入方向正确。结论成功构建了含hURAT1基因5’UTR区的荧光素酶表达载体,可用于后续功能研究。Objective To construct the lueiferase expression recombinant containing human urate transporter 1 gene （hURAT1） in 5＇untranslated region （5＇UTR）. Methods The smallest functional unit of hURAT1 gene promoter and DNA segments in 5＇UTR were amplified with the human genome DNA by PCR, the segment was cloned into the luciferase expression vector pGL3 basic. The recombinant was detected by endonuclease and sequenced. Results The sequenced segment （590 bp） obtained in the recombinants was identical to that reported by GenBank and the segment inserted in the correct direction. Conclusion The luciferase expression recombinant containing hURAT1 in 5＇UTR is successfully constructed, which can be used for subsequent function research.
