摘 要:目的了解青岛地区献血者单个核细胞(PBMC)和唾液标本中EB病毒(EBV)、人疱疹病毒6型(HHV-6)和人疱疹病毒7型(HHV-7)的感染情况。方法采用PCR-Southern blotting检测109名健康无偿献血者外周血PBMC和唾液标本中EBV DNA,巢式PCR技术检测HHV-6和HHV-7 DNA,限制性酶切分析分别对HHV-6、HHV-7进行分型和鉴定。结果109名献血者外周血PBMC中EBV DNA检出率为21.10%(23/109),HHV-6 DNA检出率为50.46%(55/109),HHV-7 DNA检出率为57.80%(63/109)。唾液标本中EBV、HHV-6和HHV-7 DNA检出率分别为24.77%(27/109)、66.06%(72/109)和90.83%(99/109)。PBMC中EBV、HHV-6和HHV-7均阳性者7例,占6.4%;唾液标本中均阳性者17例,占15.6%。酶切分析结果显示,55例献血者外周血PBMC HHV-6阳性标本中HHV-6A型占18.2%(10/55),HHV-6B型占81.8%(45/55);72例唾液HHV-6阳性标本中HHV-6A型占16.7%(12/72),HHV-6B型为83.3%(60/72)。结论外周血PBMC和口咽部上皮细胞是EBV、HHV-6和HHV-7潜伏的重要部位,献血员中存在上述病毒的共感染。Objective To investigate the prevalence of EBV, HHV 6 和 HHV 7 in peripheral blood mononuclear cells (PBMC) and saliva samples in blood donor. Methods PCR-Southern blotting was adopted to detect EBV DNA in PMBC and saliva of 109 blood donors in Qingdao~ nested PCR method to detect HHV 6 and HHV-7 DNA; restriction map to type and identify HHV-6 and HHV 7. Results Of the 109 blood donors, the detection rates of EBV DNA, HHV-6 DNA and HHV-7 DNA in PMBC were 21.10 % (23/109), 50.46% (55/109) and 57.80% (63/109), respectively; in saliva, the rates were 24. 77% (27/ 109),66.06% (72/109) and 90.83% (99/109), respectively. InPBMC, seven cases were with all the three items, EBV,HHV 6 and HHV 7, were positive, accounting for 6.4% (7/109) ; in saliva, the positive case was 17, accounting for 15. 6%(17/109). Restriction enzyme analysis revealed that 18.2% (10/55) of HHV 6A in PBMC and 16.7% (12/72) in saliva, 81.8%(45/55) of HHV 6B in PBMC and 83.3% (60/72) in saliva. Conclusion Peripheral blood mononuclear cells and epithelial cells of pharynx oralis are the important latent sites for EBV,HHV 6 and HHV 7. Co infection of EBV with HHV-6 and HHV 7 exists in blood donors.
