摘  要:考查分析了东亚飞蝗精巢细胞减数分裂各主要时期的特征及分裂高峰期,为昆虫染色体结构、功能及进化研究提供标本材料,为蝗虫染色体的后续研究奠定了基础,也为生物遗传实验教学提供一种简单易行的方法。本实验采用每只活体注射0.04%秋水仙素0.1mL、细胞悬液滴片的方法,对东亚飞蝗的精巢细胞进行染色体制片。实验观察到东亚飞蝗精巢细胞减数分裂Ⅰ、Ⅱ各主要时相的鲜明特征,双线期同源染色体的四分体结构明显,浓缩期染色体形状多种,8月中旬是制作东亚飞蝗减数分裂标本的最佳时期。Experimental observation of the meiotic process is one of the main contents in the teaching of genetics. To study the structure, function and evolution of insect chromosome, and to get more clean division phase samples, the division peak time and character of spermary cell meiosis phases of Locusta migratoria manilensis are analyzed. In this paper spermary cell chromosome specimen of L. migratoria manilensis is prepared by injecting colchicine (0.04% , 0.1 mL each) into living locust, and made the spermary to cell suspension, then dripped the suspension on slide, observed the meiotic process of chromosome clear by. The experiment indicates that it was an effective way of chromosome dispersion. In order to obtain more divided, clean and clear sample, the distinct character of spermary chromosome in different division phase is observed, the best time to make spermary chromosome samples of L. migratoria manilensi is the middle of August. It has laid a foundation to further study of grasshopper chromosome. At the same time, it offers a simple and easy experimental method for the genetics experiment teaching, and a Leaching reference for school biology.
