摘 要:目的本试验通过建立大鼠的脂肪移植模型,在移植体内加入血管生成素-1,利用血管生成素-1促进血管生长,加速局部血液循环重建的作用,来研究其对移植体成活率的影响。方法健康6个月的Wistar大鼠36只,雌性,平均体重(300±5)g。采用随机对照设计的。将所有大鼠随机分成6组,每组6只,1~2组为2μg/mL浓度的Ang-1组,3~4组为0.25μg/mL浓度的Ang-1组,5~6组为生理盐水对照组。切取大鼠双侧腹股沟处的脂肪约1 mL,纯化后按分组要求分别经2、0.25μg/mL Ang-1和生理盐水处理后,进行自体背部皮下移植。在术后30天取出移植的脂肪组织观察,测量脂肪体积,计算成活率。结果3组取出移植的脂肪组织后计算脂肪移植成活率,经高浓度Ang-1处理的移植体成活率明显高于盐水处理组的移植成活率(P<0.01),低浓度Ang-1处理组移植体成活率也高于盐水处理组的移植成活率(P<0.01),高、低浓度Ang-1处理组间差异无显著性意义。结论一定浓度的Ang-1对移植鼠颗粒脂肪组织有促进其血管增生的作用,从而提高了移植后脂肪组织存活率。Objective Through establishing a rat fat-transplantation model, injecting ang- 1 into fat particles, with ang- 1's function of improving angiogenesis and blood-circulation, to investigate its effect on survival in recipient area. Methods Thirty-six female Wistar rats weighing 295 -305 g were randomly assigned into six groups and treated as follows: both grafts in 1 st and 2nd groups were mixed with 2 μg/mL ang-1, both grafts in 3 th and 4th groups were mixed with 0. 25μg/mL ang-1, both grafts in 5 th and 6th groups were with mixed normal saline as control groups. 1 mL autogenous fat grafts were excised from bilateral inguinal areas of every Wistar rats. Then it was purified and mixed with 2 μg/mL ang-1, 0. 25 μg,/mL ang-land 0.9% NS respectively. Finally transplanted them in the rats' dorsums. Three groups' transplanted tissues were taken out after 30 days, and their volumes were measured with their survival rate being calculated. Results Microvessels should brown color after CD34 staining. The values of microvessel density in ang-lgroups was significantly higher as compared with that in saline groups in the same observing periods ( P〈0. 01 ). There was no significant differences between the two ang-1 groups. Conclusions A certain density ang-1 may be a candidate angio- genesis factor in fat particle auto- transplantation.
