摘  要:在玻碳电极上电聚合2,6-吡啶二甲醛,再共价键合硫堇至聚2,6-吡啶二甲醛膜的表面上制备成修饰电极.用电化学交流阻抗表征了该修饰电极的制备过程.以差示脉冲伏安法研究了该修饰电极对鲱鱼精DNA的作用,发现该修饰电极对dsDNA具有选择性的伏安响应.在优化的实验条件下,修饰电极氧化峰电流与dsDNA浓度的对数在0.10~100mg/L范围内呈现良好的线性关系,检测限为0.06mg/L.将该修饰电极用于检测鲱鱼精DNA的化学损伤,其峰电流与DNA的损伤程度成正比.该修饰电极制备简单、检测快速,5支电极间相对标准偏差小于4.83%.A thionine/Poly (pyridine-2,6-dicarbaldehyde) modified electrode (Th/PPDA/ GCE) was fabricated for detecting DNA and its damage. Th/PPDA/GCE was prepared by electropolymerizing pyridine-2,6-dicarbaldehyde on a glassy carbon electrode, and covalently immobilizing thionine on the surface of poly(pyridine-2,6-diearbaldehyde) film. The preparation process and electrochemical responses of Th/PPDA/GCE were investigated by using electrochemical impedance spectroscopy and differential pulse voltammetry for herring sperm DNA. The characteristic responses of Th/PPDA/ GCE for the dsDNA on the potential and current were found in favor of discrimination of the ssDNA and dsDNA. Under the optimum conditions, the current intensity was proportional to the logarithm of dsDNA concentration over the range from 0.1 to100 mg/L with a correlation coefficient of 0. 9988, and a 0.06 mg/L detection limit. The modified electrode was successfully applied to detect the oxidative damage to dsDNA mediated by Fenton reaction. The level of DNA damage can be distinctly showed by observing the change of current intensity and the shift of potential. The modified electrode also has a good stability, and the relative standard deviation is less than 4.83% for 5 electrodes.
