摘 要:以炭黑XC—72担载于PVC粉末表面所制得的XC—72/PVC复合材料为载体,利用顺序注射-紫外可见分析系统,研究了牛血清白蛋白在炭黑表面的在线吸附行为.结果表明:在pH=4.5左右,XC—72/PVC对牛血清白蛋白具有良好的吸附;在线和静态2种操作模式下的吸附行为均符合Langmuir吸附机理;相比静态吸附,在线吸附表现出操作简便、分析速度快、试剂消耗量小、自动化程度高的显著特点.认为实验结果可为研究蛋白质在炭黑表面的在线固定化提供新的技术手段和理论基础.In this work, carbon black XC-72 loaded on PVC surface was served as a novel solid-phase extraction sorbent. A SIA-UV system was developed to implement the on-line adsorption and the realtime monitoring of bovine serum albumin on the XC-72 surface. The adsorption of BSA agrees well with the Langmuir adsorption model. A thorough scrutiny of the experimental parameters affecting the adsorption of BSA from buffer solution onto the XC-72/PVC spheres was carried out. The maximum adsorption capacity of BSA was derived as 313.44 and 428.00 mg (BSA) g^-1 (XC-72) in static and flow system, respectively. The present approach has many advantages such as simple, automation, low consumption of reagent, short adsorbent time. This work can provide a new strategy for immobilization of protein and preparation of biosensors.
