摘 要:[目的]研究不同浓度氟对大鼠血清自由基和氧化应激的影响。[方法]在饮水中加入含NaF(分析纯)10,20,40mg/kg的去离子水灌胃喂饲大鼠3月后,测定尿氟、血清氟含量、血液中的钙浓度。血清中的一氧化氮(NO)、丙二醛(MDA)及谷胱甘肽过氧化物酶(GSH-Px)的含量。[结果]随染氟剂量的增加,3月后各剂量组大鼠尿氟浓度相应增加,存在剂量-效应关系(R=0.914,P﹤0.01),与对照组相比有统计学差异(P﹤0.01);各剂量组血氟浓度相应增加,存在剂量-效应关系(R=0.930,P﹤0.01),与对照组相比有统计学差异(P﹤0.05);各剂量组大鼠血钙浓度相应下降,存在剂量-效应关系(R=-0.899,P﹤0.01),与对照组相比有统计学差异(P﹤0.01);各剂量组NO的含量相应增加,存在剂量-效应关系(R=0.951,P﹤0.01),与对照组相比有统计学差异(P﹤0.01);各剂量组MDA含量升高,存在剂量-效应关系(R=0.923,P﹤0.01),与对照组相比有统计学差异(P﹤0.05);各剂量组GSH-Px的活性下降,存在剂量-效应关系(R=-0.966,P﹤0.01),与对照组相比有统计学差异(P﹤0.05),特别是高氟剂量组NO及MDA的含量最高而GSH-Px的活性最低。[结论]慢性氟中毒可导致大鼠处于氧化应激状态。[Objective] To study the effect of different dosages of fluoride in serum on the free radicals and oxidative stress of rats. [Methods] Drinking water with various concentrations of sodium fluoride was given to rats for 3 month. Then, the contents of the fluoride in the urine and serum, concentration of calcium in blood, content of nitnic oxide (NO) and maleic dialdehyde (MDA) and activities of Glutathion Peroxidase (GSH-Px) in serum were measured. [Results] With the increase of content of fluorin, the content of fluorin in urine was increased in all dosage groups after three months. Dose-response effect was existed (R=0.914, P﹤0.01), and it has significant difference when compare with the controls (P﹤0.01); The content of NO and MDA increased in all dosage groups, and dose--response effect was existed as well (NO: R= 0.951, P﹤ 0.01; MDA: R=0.923, P﹤0.01), also, it has significant difference when compare with the controls (NO: P﹤0.01; MDA: P﹤0.05). The activities of GSH-Px decreased, a dose-response effect presented, and there was significant difference compared with the control group (P﹤0.05). Especially the contents of NO and MDA was the highest while the activities of GSH-Px was the lowest in the maximal fluoride group. [Conclusion] Chronic fluorosis may induce oxidative stress to rats.
