摘  要:【目的】应用计算机辅助精子分析(CASA)系统观察昆明山海棠对大鼠精子的毒性作用。【方法】选用18只成熟雄性SD大鼠随机分为2组,每组9只:正常对照组喂服生理盐水10 mL.kg-1.d-1,中药组喂服昆明山海棠水煎液7.5 g.kg-1.d-1。连续喂服60 d后,采用Klinefelter扩散法收集大鼠附睾尾精子,采用CASA系统观察精子计数、密度、形态、活动力。【结果】与正常对照组比较,中药组大鼠附睾的精子计数和密度显著下降(P<0.01),检测视频下可见大量畸形精子,精子活动力下降为零。【结论】昆明山海棠水煎液对大鼠精子具有一定的直接毒性效应。Objective To study the toxic action of Tripterygium hypoglaucum Hutch (THH) on rat sperms by computer-assisted sperm analysis (CASA) system. Methods Eighteen male SD rats with the weight of 180 -220g were equally randomized into 2 groups: the normal control group were given normal saline in a dose of 10 mL·kg^-1·d^-1, and the experiment group were given THH water decoction in a dose of 7.5 g·kg^-1·d^-1. After treatment for 60 days, the rats were sacrificed and the sperms were isolated from the caudal epididymides of rats with Klinefeher diffusion method. Effect of THH on the motility, appearance, number and density of rat sperms was evaluated by CASA system. Results A significant decrease of the number and density of sperms was seen in the experiment group as compared with the control group (P 〈 0. 01 ). In the experiment group the motility of sperm from caudal epididymides dropped to zero, and a large amount of teratosperms were found under micro-video camera. Conclusion THH water decoction exerts certain direct toxic action on rat sperms.
