摘 要:目的通过观察氟马西尼、纳洛酮及二者合用对羟丁酸钠麻醉小鼠的催醒作用,研究羟丁酸钠催眠机制与阿片受体,苯二氮卓艹类受体的关系。方法昆明种小鼠40只分成4组(每组n=10),腹腔注射2.5 g.kg-1羟丁酸钠,待翻正反射消失后1 min,分别腹腔注射生理盐水0.1 ml.10 g-1、纳洛酮2 mg.kg-1、氟马西尼1 mg.kg-1、纳洛酮2 mg.kg-1+氟马西尼1 mg.kg-1,观察小鼠翻正反射消失的持续时间,即睡眠时间(sleepingtime,ST)。结果与生理盐水组相比,纳洛酮和氟马西尼单用组小鼠的睡眠时间有缩短的趋势,但各组之间均无统计学差异(P>0.05)。两药合用组与单独用药组差异亦无统计学意义。结论羟丁酸钠催眠机制可能与阿片受体、苯二氮卓艹受体关系不大。Objective To investigate the analeptic effects of flumazenil and naloxone administered alone and in combination on sodium oxybate-induced anesthesia in mice,so as to explore the hypnotic mechanism of sodium oxybate and the relationship between opioid receptors and benzodiazepine receptors.Methods 40 Kunming mice were randomized into 4 groups(n=10 each).Sodium oxybate(2.5 g·kg-1) was intraperitoneally administered in mice.One minute after loss of righting reflex,normal saline(0.1 ml·10 g-1),naloxone(2 mg·kg-1),flumazenil(1 mg·kg-1),and naloxone(2 mg·kg-1) plus flumazenil(1 mg·kg-1) were intraperitoneally injected.The duration of loss of righting reflex or sleeping time(ST) was recorded.Results Compared with the normal saline group,the ST in the naloxone and flumazenil groups was in the tendency of reduction,but there was no statistic difference between groups(P0.05),nor was there any significant difference between groups of the two drugs used alone and in combination.Conclusion The hypnotic mechanism of sodium oxybate has no relationship with opioid receptors and benzodiazepine receptors.
