摘 要:目的研究氯化镧(lanthanum chloride,LaCl3)对原代培养的星形胶质细胞的氧化损伤作用。方法原代培养的星形胶质细胞经0、0.25、0.5、1.0 mmol/L LaCl3处理24 h后,采用MTT法和Alamar Blue还原法检测星形胶质细胞的存活率,并测定星形胶质细胞中谷胱甘肽(glutathione,GSH)、丙二醛(malondialdehyde,MDA)含量和谷胱甘肽过氧化物酶(glutathione peroxydase,GSH-Px)活性。结果各LaCl3处理组星形胶质细胞的存活率均显著低于对照组,且具有一定的剂量-反应关系。0.25 mmol/L LaCl3组星形胶质细胞的GSH含量和GSH-Px活性显著低于对照组;0.5 mmol/L LaCl3组星形胶质细胞的GSH含量和GSH-Px活性显著低于对照组和0.25 mmol/L LaCl3组;1.0mmol/LLaCl3组星形胶质细胞的GSH含量和GSH-Px活性分别降低至对照组的66%和43%,均显著低于对照组、0.25和0.5 mmol/L LaCl3组。此外,各LaCl3组星形胶质细胞的MDA含量均显著高于对照组,且随LaCl3染毒剂量增加而增加。结论LaCl3对星形胶质细胞具有损伤作用,其机制可能与LaCl3导致星形胶质细胞氧化损伤有关。Objectives To explore the oxidative damage effect of lanthanum chloride(LaCl3)on primary cultured astrocytes.Methods Primary cultured astrocytes were exposed to 0,0.25,0.5 and 1.0 mmol/L LaCl3 respectively,for 24 h,then measure the survival rate of astrocytes(by MTT assay and Alamar Blue method),the levels of glutathione(GSH)and malondialdehyde(MDA),and the activity of glutathione peroxydase(GSH-Px)in astrocytes.Results The results showed that the survival rate,GSH level and GSH-Px activity in LaCl3 treated astrocytes were significantly lowered compared with control group,while the MDA contents in LaCl3 treated astrocytes were significantly higher than that of controls,they all were dose-dependent.Conclusion The results suggested that lanthanum chloride has damage effect on astrocytes,the mechanism may relate to oxidative damage in primary cultured astrocytes which could be induced by LaCl3,which might be an important mechanism of lanthanum chloride toxicity.
