摘  要:目的:观察补阳还五汤对血管性痴呆大鼠行为学和海马CA1区神经元形态的影响。方法:采用四血管阻断法制备大鼠血管性痴呆模型;水迷宫实验检测大鼠行为学改变;HE染色观察海马CA1区神经元的形态改变并对神经元数目进行定量分析。结果:Morris水迷宫实验结果发现,与模型组大鼠比较,补阳还五汤组、尼莫地平组大鼠的逃避潜伏期明显缩短,穿越平台区域的次数明显增多(P<0.05)。HE染色:模型组大鼠海马神经元出现明显的病理变化,补阳还五汤组、尼莫地平组大鼠海马神经元的病理改变明显改善,且补阳还五汤组、尼莫地平组大鼠海马神经元的数目明显多于模型组(P<0.05)。结论:补阳还五汤可以改善血管性痴呆大鼠的学习记忆能力,对血管性痴呆大鼠的缺血再灌注损伤有一定的保护作用。Objective:To observe the effects of Buyanghuanwu decoction on behavior and morphology of neurons in hippocampus CA1 region of vascular dementia (VD) rats,Methods:The VD rats model was made by four-vessel occlusion to observe the behavior changes with Morris water maze and the pathological changes of neurons in hippocampus CA1 region with HE staining.Results: Results of Morris water maze: compared with VD model rats, the escape latency of rats in Buyanghuanwu decoction group and nimodipine group shortened obviously and the times that rats crossing the plateau region increased obviously (P 〈 0.05). HE staining: The neurons in hippocampus CA1 region of rats in model group appeared significant pathological changes, while these pathological changes significantly improved in Buyanghuanwu decoction group and nimodipine group; moreover, Buyanghuanwu decoction and nimodipine could obviously increase the neuron numbers of model rats (P 〈 0.05).Conclusions:Buyanghuanwu decoction can improve the learning and memory ability of VD rats and protect neurons in hippocampus from ischemia-reperfusion injury.
