摘 要:目的研究半夏、钩藤乙醇提取物单用和4:1配伍对青霉素皮层定位注射诱发惊厥发作和痫性放电的作用。方法制备大鼠运动皮层定位注射青霉素诱发惊厥模型,以托吡酯(0.154g/kg)作阳性对照,半夏(36.4g/kg)、钩藤(38.5g/kg)、半夏+钩藤(24.5g/kg)灌胃(ig)后,采用RM6240C多道生理信号采集处理系统同步记录大鼠皮层和海马的EEG,观察药物对痫性行为、痫性放电潜伏期、痫波发放频率及最高波幅的影响。结果半夏、钩藤乙醇提取物单用和4:1合用,均可明显抑制痫性行为和痫性放电,其中合用组较两单药组显著延长痫性行为发作潜伏期和痫性放电的潜伏期,但对痫波发放频率及最高波幅的影响无统计学意义。结论半夏和钩藤乙醇提取物单用和4:1配伍用药能明显抑制青霉素皮层定位注射诱发大鼠惊厥和痫性放电,两药配伍应用的抗惊厥作用优于单用。Objective To study the anticonvulsant action of 4∶1 proportion compatibility of uncariae(UN)and pinellia ternate(PT) extractions on the model of seizure induced by penicillin in rats.Methods The models of seizure were induced by penicillin,topamax as the positive control drug.The model rats were randomly divided into five groups:Model group,topamax group treated by topamax,uncariae group treated by uncariae(36.4 g/kg),pinellia ternate group treated by pinellia ternate(38.5 g/kg),uncariae and pinellia ternate group treated by uncariae and pinellia ternate(24.5 g/kg).The convulsant behaviors and seizure latency,frequency of epileptiform discharge,highest wave of hippocampus and cortex EEG were observed by RM6240C multichannel physiological signal collection and analysis recorder.Results The convulsant behaviors and epileptiform discharge were inhibited in uncariae group,pinellia ternate group,uncariae and pinellia ternate group.Compaired with uncariae group and pinellia ternate group,the seizure latency and epileptiform discharge latency were prolonged in uncariae and pinellia ternate group.There was no significantly difference in reducing the frequency of epileptiform discharge and decreasing the highest wave of EEG in three groups.Conclusion The uncariae,pinellia ternate extractions and its 4:1 compatibility could effectively antagonize the seizure and epileptiform discharge induced by penicillin in rats.The anticonvulsant action of UN and PT was better than them alone.
