复方仙蓉颗粒抑制MCF-10AT乳腺癌癌前病变的研究

摘　　要：目的:建立MCF-10AT乳腺癌癌前病变模型,探讨复方仙蓉颗粒(Compound Xian Rong Granules,CXRG)对MCF-10AT乳腺癌癌前病变模型的抑制作用。方法:采用体外接种MCF-10AT细胞于BALB/c雌性裸鼠,并肌注苯甲酸雌二醇建立乳腺癌癌前病变模型。随机分为模型组、CXRG组、三苯氧胺(Tamoxifen,TAM)组,每组20只。分接种后21、35、49、63天,分组观察裸鼠移植瘤重量及体积变化,观察移植瘤组织形态学变化。结果:接种后21天起裸鼠移植瘤组织均出现不同程度的增生。接种后35～49天,3组裸鼠移植瘤组织均出现不同程度的不典型增生,发生率为80%～100%。接种后63天,共出现3例浸润癌,其中模型组2/7,TAM组1/7;而CXRG组表现与49天时相似。结论:复方仙蓉颗粒能抑制或减缓MCF-10AT乳腺癌癌前病变的增殖,作用优于三苯氧胺。Objective：To establish MCF-1OAT breast precancerous lesion model and discuss the inhibition effect ofCompound Xian Rong Granules（CXRG）to it.Methods：The MCF-10AT cell line in vitro was inoculated into the female nude mice and the breastprecancerous lesion model was induced by intramuscular injection of estradi01.Sixty nude mice were randomlydivided into three groups including the model group,CXRG group and Tamoxifen group.There were 20 nudemice in each group.The weight and volume changes of the transplanted tumor were observed separately at the 21st,35th,49th and 63rd day after inoculation,as well as the histomorphological changes.Results：the transplanted tumor tissues of the nude mice had appeared hyperplasia in different degrees since 21days after inoculation.And 35～49 days after inoculation,there were some nude mice in each group,whosetransplanted tumor tissues appeared atypical hyperplasia in different degrees,with an incidence rate of 80%～1 00%.And 63 days after inoculation,3 cases presented invasive breast cancer.2 cases appeared in themodel group,accounting for 2/7;and 1 case appeared in the txmoxifen group,accounting for 1/7.However,theCXRG group showed the same with their conditions in the 49th days after inoculation.Conclusion：Compound Xian Rong Granules can inhibit the hyperplasia ofthe MCF-10AT breast precancerous lesion and has better effect than tamoxifen.
