祛痰通络中药对大鼠脑梗死后原位干细胞增殖 分化及神经功能恢复的影响

摘　　要：目的:观察大鼠脑梗死后脑内自体神经干细胞(Neural stem cells,NSCs)的增殖、迁移与分化以及祛痰通络中药对其影响。方法:Sprague-Dawley(SD)大鼠随机分为假手术组(N组)、生理盐水组(A组)、祛痰通络中药治疗组(B组),比较各组大鼠在脑梗死模型建立后24h、3天、7天、14天各时间点神经功能缺损评分,并应用免疫组化法检测并比较各时间点自体神经干细胞增殖情况、免疫荧光双标后观察增殖干细胞的分化情况。结果:脑梗死后第14天祛痰通络中药治疗组评分低于生理盐水组(P<0.05),第3天开始增殖细胞、第7天开始分化为神经元样细胞数均多于A组(P<0.05)。结论:祛痰通络中药可促进脑梗死后自体神经干细胞的增殖并且更多的分化为神经元,从而促进脑梗死大鼠的神经功能恢复。Objective：To observed the effect of eliminating-phlegmcommunicate-collaterals decoction pieces on cerebral infarction（CI） rats,the affection on the activation,migration and differentiation of NSCs in situ after cerebral infartcion of them,and try to explore the mechanism of it.Methods：SD rats were divided randomly into 3 groups as follows：sham operation group（N）,CI group treated with saline（A）,CI group treated with eliminating-phlegmcommunicate-collaterals decoction pieces （B）. According to its neurol function,we give each rat a score at the time of 24hours,3rd,7th and 14th day after the operation,and compared with each group. The brain tissue sections were stained by Immunohistochemistry method to detect NSCs activation（BrdU＋ cells）;and we used the double immunofluorescence method to detect how many NSCs differentiated into neuron-like （BrdU＋/NeuN＋）cells,and how many gliocytes （BrdU＋/GFAP＋）cells.Results：The mean neurol function scores of B group were lower than A group on the 14th days after CI（P0.05）,from 7rd to 14th day,compared with A group,the BrdU＋/NeuN＋ cells of B group was more（P0.05）.Conclusion：eliminating-phlegmcommunicate-collaterals decoction pieces can promote the rehabilitation of CI rat ,meanwile promote self NSCs to activate and differentiate into neurons.
