生化分析仪试剂间交叉污染对总胆汁酸测定的影响及分析

摘　　要：目的探讨试剂间交叉污染对日立7600-010全自动生化分析仪测定总胆汁酸的影响。方法取混合血清,根据交叉污染项目的筛选程序筛查出对总胆汁酸测定有干扰的项目;确定总胆固醇和糖化血清蛋白试剂对总胆汁酸测定有干扰影响。分析总胆固醇试剂对胆汁酸测定的干扰影响,采取先测定总胆固醇再连续测定三次总胆汁酸的顺序,并重复十次,以确定总胆固醇对总胆汁酸的影响程度,并观察总胆固醇试剂对不同浓度胆汁酸测定的影响程度。确定总胆固醇试剂的干扰因素。应用仪器的特殊清洗程序,观察仪器特殊清洗程序排除试剂间干扰的效果。结果与单独测定TBA的对照组比较,显示总胆固醇试剂对胆汁酸测定有明显的正干扰,总胆固醇试剂对低浓度总胆汁酸的干扰较高浓度的明显。引起总胆汁酸测定结果明显升高的主要干扰试剂为总胆固醇R1试剂,采用碱液加强清洗可明显减少两者间的交叉污染。结论总胆固醇试剂对总胆汁酸的测定具有明显的正干扰,在设定分析参数时,应注意两者的顺序,并在设计生化组合套餐时,应在两者中间间隔开几个项目,或者用碱液加强清洗吸样针,避免交叉污染的产生。Objective To study the influence of the Total Cholesterol （TCHOL） reagents on the determination results of Total Bile Acids （TBA） by Hitachi 7600-010 automatic biochemical analyzer. Methods Mixed serum prepared, screened out the interference items of the determination of TBA according to the project of cross-contamination selection procedures; Defined the TCHOL which would have some influence on the determination of TBA. Determined three times of TBA after TCHOL measured, and repeated 10 times in order to determine the impact of TCHOL on TBA. Low, medium and high concentrations of TBA were determined after TCHOL to define influence among the different concentrations of TBA. Reagent 1 and 2 of TCHOL were added to mixed serum, to make sure which reagent would have cross-interference with TBA; Special cleaning procedures of equipment could exclude the effect of interference between the reagents. Results Compared with the group of determination of TBA independently, the reagents of TCHOL had a positive interference on TBA. The interference on low concentration of TBA was more obvious than that on the high one. Reagent 1 of TCHOL could significantly increase the determina- tion results of TBA, using lye to enhance cleaning agents could significantly reduce the cross-contamination of TCHOL. Conclusion reagents of TCHOL have a significant positive interference on the determination of TBA. When setting the analysis parameters, the order should be paid attention to, and when designing the biochemistry course, it should separate them with the other items, or use a special cleaning procedure with the lye cleaning between the items, to avoid cross-contamination.
