摘 要:目的对比早产胎儿脐血和脐带间充质干细胞(MSCs)的培养成功率,探讨早产胎儿MSCs获取的最佳途径。方法无菌条件下采集早产胎儿(不足37周)脐血,密度梯度离心法分离单个核细胞,采用贴壁培养法获得脐血MSCs;将脐带剪成1 mm3大小组织块,采用组织块贴壁法获得脐带MSCs;分别观察脐血和脐带MSCs的生物学特性,免疫荧光法检测MSCs表面标记物的表达情况。结果采用MesencultTM培养基,早产胎儿脐血MSCs培养成功率为100%,脐带MSCs培养成功率为67%,明显低于脐血MSCs;早产胎儿脐血和脐带MSCs均表达CD29、CD44和CD90,不表达造血干细胞表面标志CD34。结论早产胎儿脐血中较易获得MSCs,为临床移植治疗提供了理想的细胞来源。Objective To compare the achievement ratio of mesenchymal stem cells (MSCs) derived from premature fetus umbilical cord blood and umbilical cord, inorder to explore the best way of obtaining MSCs. Methods Umbilical eord blood were collected from premature fetus (less than 37 months) under aseptic condition, mononuelear cells were isolated by density gradient centrifugation, and/HSCs was obtained by attachment culture methed; umbilical cord was cut into 1 mm^3 tissue block, MSCs were separated from which with tissue attachment culture method; the biological characteristics of MSCs from umbilical cord blood and umbilical cord were observed, and the cell surface markers were detected by immunofluorescence method. Result In MeseneuhTM medium, the achievement ratio of umbilical cord blood from preterm fetus was 100% , and which of the umbilical cord was 67% (P〈0.05). MSCs from umbilical cord blood and umbilical cord in preterm fetus both expressed CD29, CD44 and CD90 abundantly, but not expressed the hematopoietie antigen CD34. Conclusion It is easy to obtain MSCs from umbilical cord blood in premature fetus, which provides ideal source for clinical cell transplantation.
