摘 要:目的研究乙型肝炎免疫球蛋白(HBIG)聚氰基丙烯酸正丁酯纳米粒(HBIG—PBCA—NP)、PBCA—NP的体外应用安全性。方法用乳化聚合法分别制备HBIG-PBCA-NP、PBCA-NP;采用MTT比色法研究不同浓度的HBIG-PBCA-NP、PBCA—NP对外周血单核细胞(PBMC)、QSG7701细胞及HepG2.2.15细胞生长抑制作用的影响;用自动生化仪测定乳酸脱氢酶(LDH)、丙氨酸转氨酶(ALT)活性。结果按相对生长率(RGR%),HBIG—PBCA—NP、PBCA—NP对PBMC、QSG7701细胞及HepG2.2.15细胞的毒性分级分别为0—1级,LDH与ALT释放值与HBIG、空白对照相比,差异无显著性(P〉0.01)。结论PBCA—NP属无毒级载体,HBIG—PBCA—NP具有良好的细胞生物安全性。Objective To investigate the application security of hepatitis B immunoglobulin polybutylcynaoacrylate nanoparticles (HBIG-PBCA-NP) and polybutylcynaoacrylate nanoparticles (PBCA-NP) in vitro. Methods HBIG-PBCA-NP and PBCA-NP were prepared respectively by emulsion polymerization method. The effects of different concentrations of HBIG-PBCA-NP, PBCA-NP and HBIG on the growth inhibition of PBMCs, QSG7701 cells, HepG 2.2.15 cells were studied by MTT colorlmetric assay; the activity of lactate dehydrogenase (LDH) and alanine aminotransferase (ALT) were detected by automatic biochemy-instrumen. Results Judging by relative growth rate ( RGR% ), the cytotoxicities grade of HBIG-PBCA-NP, PBCA-NP and HBIG for PBMCs, QSG7701 cells, HepG 2.2.15 cells were 0-1 grade, respectively. Compared to the controls,the LDH and ALT levels of HBIG-PBCA-NP, HBIG and PBCA-NP treatment groups showed no differences. Conclusion PBCA-NP is non-toxic-class carrier,HBIG-PBCA-NP has good bio-security in vitro.
