摘 要:用葫芦[Lagenaria siceraria(Molina)Stand.]作为黄瓜绿斑驳花叶病毒(Cucumber green mottle mosaic virus,CGMMV)的繁殖寄主,通过差速离心和PEG二次沉淀法进行病毒提纯,用提纯病毒液免疫新西兰兔制备CGMMV抗血清。制备的CGMMV抗血清经间接ELISA法测定效价为1∶5120;利用制备的抗血清检测田间样品,证明其可以用于CGMMV的血清学及免疫捕获RT-PCR检测。研究结果对今后开展葫芦科作物CGMMV检测,及时发现疫情以便采取防控措施,避免病毒扩散为害,确保葫芦科作物的安全生产具有重要意义。The present study has been conducted using the gourd [Lagenaria sicerada (Molina) Stand.] as propagating host of cucumber green mottle mosaic virus (CGMMV). The virus was isolated from the leaves of infected bottle gourd and purified by one cycle of differential centrifugation and double PEG precipitation. The purified virion was injected to New Zealand rabbits to produce CGMMV polyclonal antiserum. The indirect ELISA results showed that the titer of CGMMV antiserum was 1:5120. The CGMMV antiserum obtained was used to detect the field samples. The results indicated that obtained CGMMV antiserum could be used to detect serology of CGMMV and immunocapture reverse transcript PCR (ICRT-PCR). These results are important for detecting CGMMV in Curcurbitaceae plants, discovering and controlling the disease to avoid the damages caused by virus and ensure the sustaining production of Curcurbitaceae plants.
