摘 要:目的观察低剂量纳洛酮对曲马多镇痛效应的影响。方法80只昆明种小鼠随机分为两大组:40只(均为雌性)用于热板法试验,另40只(雌雄各半)用于扭体法试验。各大组再按分层随机设计法分为4组,即TS组(曲马多与生理盐水合用组)和TN1、TN2及TN3组(曲马多分别与纳洛酮1、10及100ng/kg合用组)。观察不同剂量纳洛酮对曲马多镇痛小鼠热板法痛阈(HPPT)和扭体次数的影响。结果与TS组相比,TN3组小鼠给予曲马多20min时HPPT值显著增高,差异有统计学意义的显著性(P<0.01),TN2组及TN3组扭体次数显著减少,与TS组相比差异有统计学意义(P<0.05)。结论低剂量纳洛酮(10~100ng/kg)能增强曲马多的镇痛效应。Objective To observe the effect of low-dose naloxone on analgesia of tramadol. Methods Eighty Kunming Jimpy mice were randomly divided into two groups: 40 accepted hot-plate method and another 40 accepted stretching method. Each group was randomly divided into 4 groups furtherly: TS group (tramadol combined with 0.9% sodium chloride) and TN1 group, TN2 group as well as TN3 group (tramadol combined with naloxone 1, 10 and 100 ng/kg respectively). Pain threshold in hot-plate test (HPPT) and the number of stretching times were observed. Results Compared with TS group, HPPT of Jimpy mice in TN3 group increased significantly (P 〈0.01) after 20 minutes of tramadol administration and the number of stretching times of Jimpy mice in TN2 group and TN3 group reduced significantly (P 〈0.05).
