摘  要:目的通过建立两次打击大鼠致多器官功能障碍综合征(MODS)的动物模型,探讨黄芪注射液对MODS致心肌和血管内皮细胞损伤的影响。方法将92只SD大鼠随机分为对照组、MODS组和治疗组。MODS组和治疗组复制大鼠MODS的动物模型,治疗组腹腔注入黄芪注射液,对照组和MODS组注入0.9%氯化钠溶液。分别在注射黄芪注射液和0.9%氯化钠溶液后24h,48h采集血液和心脏组织标本,测定大鼠血清中循环内皮细胞(CEC)数量、一氧化氮(NO)、内皮素-1(ET-1)、肌酸激酶同工酶(CK-MB)、肌酸激酶(CK)、乳酸脱氢酶(LDH)、心肌组织中肿瘤坏死因子-α(TNF-α)和细胞间黏附分子(ICAM-1)的水平,心脏组织标本行光镜下病理检查。结果治疗组大鼠腹腔注射黄芪注射液后24h和48hCEC数量、NO、ET-1水平明显低于相同时间点MODS组,差异有统计学意义(P<0.05);治疗组大鼠腹腔注射黄芪注射液后24h和48hTNF-α、ICAM-1、CK-MB、CK、LDH水平明显低于MODS组相同时间点水平,差异有统计学意义(P<0.05)。结论黄芪注射液对两次打击致MODS模型大鼠的心肌和血管内皮细胞损伤有明显的减轻和保护作用。Objective To study the effect of astragalus injection on traumatic myocardium and vascular endothelial cells in rat models of multiple organ dysfunction syndrome(MODS) induced by a harmful two-hit.Methods Ninety-two SD rats were randomly divided into control group (injected with 0.9% normal saline),MODS group (MODS rats injected with 0.9% normal saline),and astragalus group (MODS rats injected with astragalus).Blood and heart tissue specimens were colleted at 24h and 48h after injection.The amount of circulating endothelial cells (CEC) and serum levels of nitrogen monoxidum (NO),endothelin-1 (ET-1),isoenzymes of creatine kinase (CK-MB),creatine kinase (CK),lactic dehydrogenase (LDH),tumor necrosis factor-α (TNF-α) and intercellular adhesion molecule-1 (ICAM-1) were determined.Pathological examinations were performed on the heart tissue specimens using light microscopy.Results The amount of CEC and the serum levels of NO and ET-1 were significantly lower in astragalus group than in MODS group and control group at 24h and 48h (P〈0.05).The serum levels of TNF-α,ICAM-1,CK-MB,CK,and LDH were extremely significantly lower in astragalus group than in MODS group and control group at 24h and 48h (P〈0.01).Conclusion Astragalus can protect and reduce myocardium and vascular endothelial cell injury in rat models of MODS induced by a harmful two-hit.
