摘 要:目的研究慢性丙型肝炎患者外周血单个核细胞(peripheralbloodmononuclearcel,PBMC)中丙型肝炎病毒的定位分布及复制。方法直接原位反转录-PCR(标记引物)对20例慢性丙型肝炎患者的PBMC内丙型肝炎病毒(HCV)的分布、定位及存在形式进行研究。结果16例患者外周血单个核细胞的胞浆内发现HCVRNA阳性信号,并有一例患者的胞浆内发现HCVRNA负链信号。结论HCV可感染PBMC并在其中复制。原位RT-PCR为进一步研究HCV进入人体内的分布、潜伏、复制状况以及丙型肝炎治疗、药物研究奠定了基础。Objective To detect the localization, distribution, replication of HCV in PBMCs of patients with chronic hepatitis. Method Direct in situ RT-PCR with labelled primer. Results In 20 patients, 16(80%) were found positive for HCV genome within their PBMCs by in situ RT-PCR, and 1 patient was detected HCV RNA negative strand. These positive signals were localized in cytoplasma of PBMCs. Conclusion PBMCs could be an important extrahepatic site of virus replication. The destribution, localization, latent replication of HCV in bodies of patients and effect of anti-viral drug might be studied by in situ RT-PCR. HCV persisted in PBMCs can interfere with the immunologic mechanism of the patient and play a role in the pathogenic progress.
