摘 要:采用原子力显微镜(AFM)对改性前后材料表面粘附生长的角膜基质细胞的亲和力、三维形貌和力学性能进行了分析。结果显示,改性后材料的细胞亲和力有较大改善,且材料表面细胞三维形态更加正常,铺展更为舒展,与材料的接触面积较大。细胞力学性能分析也发现改性后材料表面细胞具有更高的粘附力和杨氏模量,以及更低的硬度,说明未改性p(HEMA-MMA)材料具有明显的细胞毒性,这种毒性作用导致在其表面生长的细胞的细胞骨架遭到破坏,细胞健康状态明显不如改性后材料表面细胞。因此,改性后的Col/bFGF-p(HEMA-MMA)更适合作为人工角膜材料使用。The attachment, morphology and biomechanical changes of keratocytes cultured on p( HE- MA -MMA) and p( HEMA -MMA) modified by collagen and basic fibroblast growth factor( Col/bF- GF- p(HEMA- MMA)) was studied by cell adhesion assay and atomic force microscopy(AFM). The results of cell adhesion assay showed that the attachment of keratocytes on the modified membrane was stronger than that on the unmodified membrane. The cell cultured on modified membrane presented a completely elongated and spindle-shape morphology. The biomechanical analysis of keratocytes indicated that the cell cultured on the modified membrane has higher adhesion force (2 328 ± 523 pN) and Young's modulus(0. 51 ±0. 125 kPa), and lower stiffness(0.08 ±0. 022 mN/m) than those on the unmodified membrane. The unmodified membrane had stronger toxicity on the cell cultured on its surface by damaging the cytoskeleton and health state of the cell. Therefore, the modified Col/bFGF -p( HEMA -MMA) may hold great promise to be an appropriate material for artificial cornea.
