摘 要:目的探讨宫内缺氧对幼年大鼠海马CA3区神经元与学习能力的影响,以及NMDAR1 mRNA在其新生期的表达与当归的调控作用。方法健康SD孕鼠30只,随机分为对照组、缺氧组和当归组各10只,于孕14 d开始将缺氧组与当归组孕鼠用三气培养箱制作胎鼠宫内缺氧模型,当归组用250 g/L当归静脉注射干预。3组孕鼠分娩当日每窝随机选取新生鼠2只,取脑、常规石蜡切片;余新生鼠随机选取2只喂养至30 d进行Morris水迷宫测试,测试结束当日,多聚甲醛灌注固定取脑、常规石蜡切片。幼鼠脑组织作NSE mRNA原位杂交、新生鼠脑组织作NMDAR1 mRNA原位杂交。结果Morris水迷宫实验和NSE mRNA原位杂显示:与对照组(n=20)相比,缺氧组幼鼠(n=20)空间探查测试和对位探查测试时间较缩短、海马CA3区阳性细胞数减少(P<0.05),当归组(n=20)探查测试时间较缺氧组延长(P<0.05)、海马CA3区阳性细胞数增多(P<0.05);NMDAR1 mRNA原位杂交显示:与对照组相比,缺氧组海马CA3区阳性细胞积分光密度值增大,当归组较缺氧组减小(P<0.05)。结论宫内缺氧可致新生大鼠海马CA3区NMDAR1 mRNA表达增高,从而使幼年大鼠该区神经元减少,学习记忆能力降低,而当归注射液可改善宫内缺氧状态,增加幼年大鼠海马CA3区神经元数量,提高其学习记忆能力。Objective To explore the effect of intrauterine hypoxia on hippocampal CA3 area neurons and learning capacity of juvenile rats,and to explore the expression of N-methy-D-aspartate receptor-1(NMDAR1) in hippocampal CA3 area of neonatal rats following intrauterine hypoxia and the regulatory mechanism of Angelica sinensis.Methods Thirty healthy pregnant SD rats were divided randomly into control group,hypoxia group and Angelica group.Hypoxia group and Angelica group pregnant rats were made the intrauterine hypoxia model of fetal rats with the three-gas incubator from the beginning of pregnant 14th day,and the rats of Angelica group were intervened with Angelica sinensis injection.After birth,two neonatal rats were selected randomly from each litter to take the brain tissue,make paraffin sections conventionly.Two neonatal rats were selected again randomly from the others in each litter to perform of Morris water maze after the 30th day of birth,and perfused paraformaldehyde at the end of the last day,took the brain tissue of juvenile rats,and paraffin sections conventionly.The expression of NSE mRNA and NMDAR1 mRNA were detected by using in site hybridization.Results In hypoxia group juvenile rats(n=20),the searching time of probe trial and reveral probe trial in the target quadrant,and the quantity of positive NSE mRNA cells in hyppocampal CA3 area were less than that of control group(n=20)(P〈0.05);and that of Angelica group(n=20) was more than that of hypoxia group(P〈0.05).The integral optical density(IOD) value of positive NMDAR1 mRNA cells in hippocampal CA3 area in hypoxia group neontal rats(n=20) was significantly higher than that of control group(n=20) and also higher than that of Angelica group(n=20)(P〈0.05).Conclusion Intrauterine hypoxia can increase the expression of NMDAR1 mRNA in hippocampal dentate gyrus of neontal rats,thus reduced the quantity of hippocampal dentate gyrus neurons and learning-memory capacity of juvenile rats,while Angelica sinensis inj
