摘  要:目的观察重组人生长激素(recombinant human growth hormone,rhGH)对大鼠脑缺血/再灌注损伤后缺血侧大脑皮质存活素(Survivin)和半胱氨酰天冬氨酸特异性蛋白酶-3(Caspase-3)表达的影响。方法采用大鼠局灶性脑缺血/再灌注损伤模型,将72只健康雄性SD大鼠,随机分为假手术组、对照组和治疗组。分别于模型制作成功后1、3、5、7 d处死大鼠,采用连续切片免疫组织化学技术分别检测缺血侧大脑皮质Survivin和Caspase-3的表达。结果治疗组Survivin表达较对照组明显增多,而Caspase-3表达较对照组显著减少。结论脑缺血/再灌注损伤早期应用rhGH可上调Survivin表达及下调Caspase-3表达,抑制神经元的凋亡。Objective To observe the effects of recombinant human growth hormone(rhGH) on the expressions of Survivin and Caspase-3 in rats with cerebral ischemia-reperfusion injury.Methods The model of focal cerebral ischemia-reperfusion injury was made by occluding middle cerebral artery(MCA)for 2 hours and reperfusion.Seventy-two healthy male SD rats were randomly divided into the sham-operated group,control group and treatment group.And immunohistochemistry technique was used to detect the condition of the expression of Survivin and Caspase-3 in three groups respectively.Results The expression of Survivin was markly increased and Caspase-3 was obviously decreased in treatment group compared with control group.Conclusion It is demonstrated that treatment with recombinant human growth hormone could induce expression of Survivin and suppress the activity of Caspase-3 in neurons within the cerebral cortex.It can inhibit the neuronal apoptosis.
