摘  要:目的:探讨耐力运动和补充益气补肾中药对大鼠血清和睾丸睾酮水平以及骨骼肌AR mRNA表达的的影响。方法:50只Wistar大鼠随机分为安静对照组(n=10)、安静服药组(n=10)、训练组(n=15)和训练服药组(n=15)。经6周递增负荷游泳训练后,使用放免法测定大鼠睾丸和股四头肌匀浆总睾酮水平,利用RT-PCR方法检测大鼠深层股四头肌AR mRNA的表达水平。采用One-way Anova对组间数据进行比较,采用析因方差分析揭示训练和中药两种因素对所测指标的影响。结果:未服药训练大鼠的血清和睾丸组织睾酮水平明显下降(P<0.05);服用中药的运动大鼠血清和睾丸睾酮浓度未下降。训练对大鼠骨骼肌AR基因的表达有极显著的影响(p<0.001);中药同训练因素一样,亦能明显提高AR基因的表达水平,但是效应程度低于训练因素(p<0.01);两者的交互作用对AR基因表达的影响同样显著(p<0.01)。结论:长时间适应性训练和补充益气补肾中药可以促进增强大鼠股四头肌(Ⅱ型肌纤维为主)AR mRNA的表达。The Objective of the study is to explore the effects of endurance training and the use of Chinese Medicine Compound for Invigorating Qi and Tonifying the Kidney on AR expression level of skeletal muscle in rats.Methods: 50 Wistar male rats were randomly divided into 4 groups: sedentary group(S,n=10),sedentary with medicine group(SM,n=10),training group(T,n=15) and training with medicine group(TM,n=15).After 6 weeks swimming training,testicular and muscular(quadriceps femoris) homogenate testosterone level of rats were tested through radioimmunological method;using RT-PCR,the muscular AR mRNA levels of deep quadriceps femoris in rats was measured.Results: 1) There were no significant differences of total protein content from muscle samples among every group,but there was a decrease tendency in training rats,especially in T group,10.2% and 11.9% less than that of S and SM group respectively.2) After training,testicular testosterone concentration in T group was markedly lower than that in SM and TM groups separately(P0.05).3) Compared with S group,the muscular expression of AR mRNA was obviously increased 30.3%,91.4% and 100% than that in SM,T and TM group respectively(P0.001),and 4) the ratios of AR/β-actin mRNA in quadriceps femoris of T and TM groups were increased evidently 51.2%(P0.001) and 53.5%(P0.001) than that in SM group respectively.Conclusion: 1) The results from data indicated that AR gene expressions at mRNA level could be enhanced through long-term strenuous training.2) Chinese Medicine Compound for Invigorating Qi and Tonifying the Kidney also could help increase the AR mRNA expression in skeletal muscle.
