摘 要:目的:观察有氧运动对糖尿病肾损伤大鼠肾功能保护作用,并探讨有氧运动改善糖尿病大鼠肾病的作用机制。方法:建立糖尿病大鼠肾损伤模型和有氧运动模型。比较大鼠血糖、肾脏肥大指数(肾重/体重)、尿素氮(urea nitrogen,BUN)、血肌酐(Cr)2、4 h尿蛋白(Urine protein 24 hours;UP24)等肾功能相关指标,比较各组血清超氧化物歧化酶(Superoxide dismutas,SOD)活性、脂质过氧化代谢产物丙二醛(malondialdehyde,MDA)含量、肾组织醛糖还原酶(aldose reductase,AR)、转化生长因子β1(Transforming growth factor-β1,TGF-β1)和Smad表达的改变。结果:有氧运动组与模型对照组相比,能明显降低血糖、有效控制体重下降、明显改善肾脏肥大指数、BUN、Cr、UP24、MDA含量显著降低(P<0.05),血清SOD活性增强,肾脏AR mR-NA、TGF-β1 mRNA和Smad蛋白表达水平下调(P<0.05)。结论:有氧运动对糖尿病肾损伤大鼠可能通过提高机体抗氧化能力、减少肾脏AR基因表达以抑制多元醇通路的激活、减少TGF-β1 mRNA和Smad蛋白表达以抑制TGF-β1/Smad信号通路等方面发挥其肾功能的保护作用。Objective:To investigate the protective effect of aerobic exercises on renal function and its possible mechanisms on rats with diabetic nephropathy(DN).Methods: Diabetes is induced by intraperitonal injection with streptozotocin(STZ) in male Wistar rats.The fasting blood glucose(FBG),blood urea nitrogen(BUN),serum creatinine(Cr) and urine protein 24h(UP 24 h) are measured on commercially used kits.Meanwhile,superoxide dismutase(SOD) activity,malondialdehyde(MDA) content in serum,aldose reductase(AR) mRNA,TGF-β1 mRNA and SmadProtein expression in kidney are detected by different methods.Results: The treatment of berberine significantly ameliorated the above indices including FBG,body weight,KW /BW,BUN,Cr and UP 24h compared with model group(P0.05). Berberine significantly increases SOD activity and decreases MDA content in serum compared with those in model group(P0.05).mRNA,TGF-β1 mRNA and Smad protein expression in kidney is remarkably decreased in berberine group compared with the model group(P0.05).Conclusions: These results suggested that aerobic exercise can ameliorate renal dysfunction of rats with DN through blood glucose control,reduction of oxidative stress,inhibition of the activation of polyol pathway and inhibition of the activation of TGF-β1/Smad pathway.
